I would say do not miss the 'old town' streets. The canal houses and typical historic Amsterdam scenery. You can comfortably walk the whole route. About 2 hours' walk. Ask for a map at your hotel to see at which point to start, closest to your hotel. The canal boats also depart from a few points - you can find that on the map. The normal 'sightseeing' canal ride takes about 1 hour. You will see most of the same that you have seen on your walk, but it is nice to listen to the history. If you prefer going by boat only, you can take the hop-on hop-off canal boats. They work the same as the buses - get off where you want to spend some time. Enjoy your visit, Amsterdam is a city with it's own unique charm! (And watch out for bicycles ;)
